O-GlcNAc transferase (OGT) glycosylates a large and growing list of human proteins. Here, the authors identify a surprisingly large number putative target proteins using azo-GlcNAc (GlcNAz), on-resin Click chemistry, and selective proteolysis. Notably, they validate a small subset of these by identifying specific sites of O-GlcNAcylation. While more work is needed to validate low-abundance targets, this study provides an unbiased, top-down analysis of the effects of OGT regulation in human cells.
