Temperature has significant influence on survival, growth and physiology of all living organisms. Apart from the majority of species, which growth optimally in a range of 20-40 °C, some species are able to survive in extreme conditions, such as hot springs or icy Antarctic waters. The major limitation for those organisms is the temperature, where their crucial proteins denature or become inactive.
Our mass-spectrometry based proteomics approach allow us to quantitatively monitor protein stability across broad ranges of temperature in proteome-wide scale. We employ this powerful tool to determine factors influencing protein thermal stability. This project focuses on answering the questions why certain proteins extracted from organisms living in various environmental niches melt in different temperatures and what are the factors influencing thermostability of those proteins.
