Organic synthesis of small molecule protein inhibitors
To obtain new small molecule inhibitors or immobilizable affinity probes, we use the diversity of organic chemistry reactions classically found in medicinal chemistry. In particular, palladium catalysed cross-couplings (Suzuki, Sonogashira, …) are widely used to assemble (hetero)aromatic building blocks.
Our organic chemistry laboratory is equipped with a microwave reactor (Anton Paar Monowave 300), an automated flash purification system PuriFlash 430evo (Interchim) with multi-wavelength detector, and an analytical LC-MS/MS system comprising an Agilent 1100 HPLC with autosampler and a diode array detector coupled to a Bruker amaZon ETD ion trap mass spectrometer. We have routine access to the NMR (300, 400, 500 MHz and 500 cryo) and elemental analysis facilities of the university.
